Overview
To express Cal-light TM-CaM-NES-TEV-N-AsLOV2-TEVseq-tTA controlled by light and calcium ions.
TEVp is split into N-terminal and C-terminal fragments and inserted into CaM and M13, respectively. Only when the concentration of intracellular Ca2+ increased, M13 would bindTo CaMTo make the N-terminal and C-terminal of TEV closeTo obtain proteasome activity. Blue light irradiation could expose TEVseq, thereby cutting TEV and releasing tTA. The released tTA enters the nucleusTo activate the expression of TetO promoter, thus achieving the goal of gene expression controlled by light and calcium ions.
